Abstract
Rod Enterococcus je izolirani patogen i često je uzročnik infekcija kod ljudi. Dio su normalne mikrobiote probavnog sustava te kod ljudi su najčešće izolirane vrste E. faecalis i E. faecium. Najčešće su uzročnici bolničkih infekcija, a osobito u jedinicama intenzivne skrbi. Uzrokuju infekcije urinarnog trakta, kiruške infekcije na mjestu zahvata te u nekim slučajevima može uzrokovati bakterijemiju i menigits. Cilj ovog rada bio je ispitati interakciju različitih antibiotika na adheziju uropatogenih izolata enterokoka. U svrhu ovog istraživanja koristili su se uropatogeni sojevi E. faecalis. Pomoću metode šahovnice ispitala se potencijalna sinergija dva antibiotika nitroksolin i gentamicin. Također, provedeno je inhibicijsko djelovanje dva antibiotika na adheziju. Ispitivani antibiotici, nitroksolin i gentamicin u kombinaciji pokazuju sinergistički, aditivni, indiferentni učinak. Dobiveni rezultati su pokazali da je dobiven puno veći efekt kombinacijom dvaju antibiotika manjih koncentracija. Istovremeno kombinacija antibiotika pokazuje i antiadhezijski učinak u nižoj koncentraciji od pojedinačnih antibiotika. Zaključno, kombinacija nitroksolina i gentamicina pokazuje veliki potencijal za primjenu u terapiji kroz antimikrobni i antiadhezijski učinak.
Abstract (english)
The genus Enterococcus is an isolated pathogen and is often the cause of human infections. They are part of the normal microbiota of the digestive system, and the most frequently isolated species in humans are E. faecalis and E. faecium. They are the most common cause of hospital infections, especially in intensive care units. They cause urinary tract infections, surgical infections at the procedure site, and in some cases can cause bacteremia and meningitis. The aim of this work was to examine the interaction of different antibiotics on the adhesion of uropathogenic isolates of enterococci. For the purpose of this research, uropathogenic strains of E. faecalis were used. Using the checkerboard method, the potential synergy of the two antibiotics nitroxoline and gentamicin was tested. Also, the inhibitory action of two antibiotics on adhesion was performed. The tested antibiotics, nitroxolin and gentamicin in combination show a synergistic, additive, indifferent effect. The obtained results showed that a much greater effect was obtained by combining two antibiotics of lower concentrations. At the same time, the combination of antibiotics also shows an anti-adhesion effect in a lower concentration than individual antibiotics. In conclusion, the combination of nitroxoline and gentamicin shows great potential for use in therapy through antimicrobial and antiadhesion effects.
