Abstract
Ciklus stanične diobe omogućuje povećanje mase i volumena stanice. Pogreške koje nastaju zbog poremećaja mehanizama koji reguliraju rast stanice dovode do patoloških stanja koja uključuju metaboličke poremećaje, upalne procese i zloćudnu preobrazbu stanica. Prolazak stanice kroz stanični ciklus reguliran je brojnim izvanstaničnim i unutarstaničnim signalima kao i regulatornim molekulama. Jedna od regulatornih molekula tog složenog procesa je protein Cdc6. Protein Cdc6 ima ključnu ulogu u regulaciji početnih koraka udvajanja DNA. Pogreške koje nastaju tijekom udvajanja DNA dovode do genomske nestabilnosti i zloćudne preobrazbe stanice. Brojna istraživanja pokazala su da je izražaj proteina Cdc6 povećan u različitim vrstama tumora. Stoga, za bolje razumijevanje molekularnih mehanizama putem kojih Cdc6 sudjeluje u regulaciji staničnog ciklusa, potrebna su kvalitetna protutijela. U ovom radu testirala sam primjenjivost monoklonskog protutijela protiv proteina Cdc6 u western blot analizi i ELISA analizi. Rezultati pokazuju da protutijelo specifično prepoznaje protein Cdc6 u western blot analizi i ELISA analizi. Osim toga, u budućnosti bi trebalo testirati primjenu ovog protutijela i u drugim metodama.

The cell cycle leads to an increase in mass and volume of cells. Errors that occur from disorders of regulating mechanisms of cell growth lead to pathological conditions including metabolic disorders, inflammation, and malignant transformation of cells. The passage of cells through the cell cycle is regulated by a number of extracellular and intracellular signals as well as regulatory molecules. One of regulatory molecules of that complex process is Cdc6 protein. The Cdc6 protein has a key role in the regulation of the initial steps of DNA duplications. Errors that occur during the duplication of DNA lead to genomic instability and malignant transformation of cells. Numerous studies have shown that the increased expression level of Cdc6 protein is in cells of different cancer types. The antibodies of great quality are required for better understanding the molecular mechanisms by which Cdc6 participates in cell cycle regulation. In this work, I tested the applicability of monoclonal antibodies against the Cdc6 protein in western blot analysis and ELISA analysis. The results show that the antibody specifically recognizes Cdc6 protein in western blot analysis and ELISA analysis. In addition, in the future this antibody should be tested in other methods as well.
