Abstract
Direktiva o kakvoći vode za kupanje (2006/7/EZ) za detekciju Escherichie coli propisuje dvije referentne metode: ISO-9308-1 i ISO 9308-3 (1). Revidirana je ISO 9308-1 metoda stupila na snagu 2014 g., a temelji se na korištenju kromogenog koliformnog agara. Zbog niske selektivnosti navedenog agara, ISO je preporučio primjenu metode primarno za vode s niskim brojem bakterija. Uzimajući u obzir produljeno vrijeme koje je potrebno za analizu (do 72h) i nedostatak iskustva u primjeni ISO 9308-3 u Sredozemnom moru, procjenjena je prikladnost ISO 9308-1 za ispitivanje E. coli u morskoj vodi. Cilj ovog rada bila je usporedba podataka dobivenih referentnom metodom u uzorcima morske vode prikupljanim na 110 plaža (N = 477) s podacima dobivenim primjenom šest alternativnih metoda. ISO 9308-1 – modificirana tempetarura (44 °C) značajno ne mijenja rezultate, međutim uvelike olakšava očitavanje ploča, dopuštajući korištenje dotične metode za monitoring obalnog mora.

Bathing Water Directive (2006/7/EC) for Escherichia coli detection specifies two reference methods: ISO 9308-1 and ISO 9308-3. The revised ISO 9308-1 has come into force in 2014, based upon the use of Chromogenic Coliform Agar (CCA). Due to low selectivity of CCA agar, ISO recommended this method primarily for water with low numbers of bacteria. Taking into account extended time needed for the analysis (up to 72 h) and generally lack of experience using ISO 9308-3 in Mediterranean water, suitability of ISO 9308-1 for examination of E. coli in bathing water was evaluated. Present paper was aimed to compare data obtained with a reference method in marine water samples collected at 110 beaches (N = 477) with data of the six alternative methods. Temperature modified ISO 9308-1 (44 °C) does not significantly alter the results, but greatly simplifies reading of the plates, allowing use of respective method for monitoring of coastal water.
