Abstract
Nealkoholni steatohepatits je najčešća bolest jetre. Važnost u proučavanju mehanizama nastanka ove bolesti leži u rastućoj prevalenciji, mogućoj malignoj progresiji i nedostatnoj terapiji. Ključni patofiziološki događaji u nastanku NASH-a su steatoza i lipotoksično djelovanje masnih kiselina, oksidativni stres koji oštećuje hepatocite, povećana propusnost crijevne barijere, upala koju čine pridošle stanice urođenog i stečenog imuniteta te odlaganje kolagena koje dovodi do fibroze. U progresiji bolesti najvažniju ulogu imaju γδ limfociti T u miševa, što najvjerojatnije odgovara MAIT stanicama u čovjeka. Obje vrste stanica stvaraju citokin IL-17A čiji udio u jetri tijekom vremena progresivno raste, a čije je djelovanje dominantno poticajno na nastanak upale i fibroze.
Abstract (english)
Nonalcoholic steatohepatitis is the most common liver disease. The importance in studying mechanisms of this disease lies in the growing prevalence, possible malignant progression and insufficient therapy. The key pathophysiological events in the development of NASH are steatosis and lipo-toxicity of fatty acids, oxidative stress that damages hepatocytes, increased permeability of intestinal barriers, inflammation made by innate and adaptive immune cells and collagen deposition leading to fibrosis. The most important role in disease progression is played by γδT cells in mice, which most likely corresponds to MAIT cells in humans. Both cell types produce cytokine IL-17A that increases progressively over time and acts predominantly negative on the development of inflammation and fibrosis.
