Abstract
Cilj istraživanja: Glavni cilj ovog istraţivanja bio je ispitati ekspresiju LIGHT- HVEM ligand - receptorskog para TNF superobitelji u lichen planusu, upalnoj, Th1 posredovanoj, bolesti koţe. Uloga ovih molekula uočena je u raznim upalnim procesima, kao što su upala sluznice pluća i crijeva, artritisa, inzulin-ovisnog dijabetesa i bakterijskih infekcija. Materijal i metode: Imunohistokemijsko ispitivanje HVEM i LIGHT molekula provedeno je na bioptatima lezijske i klinički nepromijenjene koţe bolesnika s lichen planusom te na uzorcima zdrave koţe (n = 10 po skupini). Fenotip stanica koje eksprimiraju HVEM receptor dodatno je ispitan metodom dvostruke imunofluorescencije i kolokalizacije s površinskim staničnim biljezima CD4, CD8 i CD68. Rezultati: Imunohistokemijsko obojenje za HVEM i LIGHT detektirano je u epidermisu i dermisu. Intenzitet obojenja znatno je jači u lezijski promijenjenoj koţi u usporedbi s nepromijenjenom i zdravom koţom. HVEM receptor jače je izraţen u niţim slojevima epidermisa, a nazočan je meĎu svim ispitanim subpopulacijama stanica upalnog infiltrata. Zaključak: Povećana epidermalna ekspresija LIGHT i HVEM u lichen planusu, kao i njegova prisutnost na raznim stanicama dermalnog upalnog infiltrata ukazuju na moguću ulogu LIGHT-HVEM signalizacije u patogenezi ove upalne bolesti koţe. Stoga rezultati ovog rada otvaraju novo poglavlje za buduća istraţivanja.

Objectives: The main goal in this study was to investigate the expression of TNF superfamily members receptor HVEM and its ligand LIGHT in lichen planus, an inflammatory disease of the skin characterized by Th1 response. The role of this ligad-receptor pair has been recognized in various inflammatory settings such as mucosal inflammation in the lung and gut, arthritis, insulin-dependent diabetes and bacterial infections. Material and Methods: Immunohistochemistry for HVEM and LIGHT was performed on biopsies of lesional and non-lesional skin of patients with lichen planus and on samples of healthy skin (n=10/group). Phenotype of cells that express HVEM receptor was additionaly examined by means of double immunofluorescence staining and analysis of colocalization with cell surface markers CD4, CD8 and CD68. Results: Immunohistochemical staining for HVEM and LIGHT was detected both in the epidermis and dermis. In lichen planus, staining was significantly stronger in lesional than in the corresponding non-lesional or haelthy skin. Staining for HVEM receptor was stronger in lower layers of the lesional epidermis. HVEM was detected among all examined populations of inflammatory cells. Conclusion: Increased epidermal expression of LIGHT and HVEM in lichen planus as well as its presence on various cells of the inflammatory infiltrate indicate a possible role of LIGHT-HVEM-mediated signaling in the pathogenesis of this inflammatory skin disease and open a new area for future research.
